Our understandings on the detailed enzyme chemistry have been greatly improved over the past several decades, mainly due to the development and application of a variety of scientific disciplines among which reaction kinetics and structural chemistry deserve particular notions for their crucial roles. Organic synthesis has been also extensively employed as an essential strategy to address numerous problems, especially refractory to be approached by other means. The scope of organic synthesis in investigation of enzyme chemistry is not confined to mere supply of substrate molecules, but is rather extended to design and application of substrate analogues as mechanistic probes to provide unique opportunities enabling analysis of enzyme catalysis in unprecedented details. In this presentation, a number of examples how organic synthesis is applied to study enzyme functions and mechanisms will be discussed and thereby the power of organic synthesis in the exploration of unusual enzyme chemistry will be demonstrated.
