Role of mitochondrial Sir2 proteins in metabolism and aging

-Abstract-
Sir2 is an NAD-dependent deacetylase that connects metabolism with longevity in yeast, flies, and worms. Mammals contain seven homologs of yeast Sir2 (SIRT1-7), which regulate multiple age-associated diseases, including cancer, diabetes and neurodegeneration. Three sirtuins (SIRT3-5) reside in the mitochondria and play a role in coordinating mitochondrial metabolism to facilitate nutrient adaptation. I will discuss the roles of these sirtuins in mitochondrial energy production.
